You need to understand the Excel object model..

If you dont plan on using a third party tool that abstracts the process of dealing with Excel, you need to get your head around the Excel object model. Assuming you are using Excel 2007, this is a good place to start => http://msdn.microsoft.com/en-us/library/bb149067.aspx. There will also be a mountain of tutorials on the web to help you as well.
